Abstract

The gene coding for -galactosidase in was detected in a recombinant gene library constructed in phage . The gene was subcloned into plasmid vectors and shown to specify a novel protein of 80000. Characterization of -galactosidase from and from recombinant expressing indicated that the enzyme functions as a tetramer. The amino acid composition of the -galactosidase, deduced from nucleotide sequencing of , gave a predicted of 82022 and revealed regions of homology to -galactosidases encoded by the Raf plasmids and by . Inactivation of the gene in resulted in loss of all -galactosidase activity and abolished the ability to ferment melibiose; -glucosidase activity was also lost, due to an indirect effect on the gene.
